Lion's mane (Hericium erinaceus) is an edible mushroom studied mainly for the brain. What is lion's mane?

It stands apart from the rest of the category because its research interest is overwhelmingly neurological. Where reishi is studied for stress and cordyceps for energy, lion's mane is studied for the brain and, increasingly, for the connection between the gut and the brain.
A short history of lion's mane
Lion's mane has a long record in East Asian traditional medicine. In Traditional Chinese Medicine it is known as hóu tóu gū, or "monkey head mushroom," and was used as a tonic for the stomach and digestive system and to support general vitality.
In Japanese herbal tradition it is called yamabushitake, after the yamabushi mountain ascetics who reportedly used it and associated it with concentration and mental clarity. That old association with focus foreshadows the modern scientific interest in its effect on the brain.
Alongside its medicinal reputation, lion's mane was prized as a culinary delicacy in imperial Chinese cuisine. This dual role as both food and medicine helped preserve its use across generations. Modern cultivation has since made it widely available in capsules, powders, coffees and nootropic blends, which is what triggered the current wave of research.
The compounds researchers focus on
Lion's mane contains a wide range of molecules, but the neurological interest comes down to two families, supported by a third.
Hericenones and erinacines
These compounds are largely unique to Hericium species. Hericenones are found mainly in the fruiting body and erinacines in the mycelium. Both have been shown to stimulate the synthesis of nerve growth factor in laboratory and animal studies (Kawagishi et al., 2008). Notably, erinacine A and erinacine S have been shown to cross the blood-brain barrier in animal models, which matters because many compounds cannot reach the brain directly.
Polysaccharides and beta-glucans
These are studied for immunomodulatory, antioxidant and anti-inflammatory activity in preclinical models (Zhu et al., 2015). They are also of interest for how they may influence gut microbiota and the intestinal barrier, which connects to the gut-brain research below.

The thread tying the neurological interest together is nerve growth factor, or NGF, and the related brain-derived neurotrophic factor, or BDNF. Both are proteins involved in the survival, growth and plasticity of neurons. NGF is studied for its role in neurogenesis, synaptic plasticity and myelination. Lion's mane compounds are researched for their ability to influence these pathways, which is the mechanistic reason for the interest in cognition.
What the research is exploring
Honesty matters most here, because lion's mane is heavily over-marketed. The mechanism research is genuinely interesting. The human evidence is early and mixed.
Cognition in older adults
The most cited human trial involved a small group of older adults with mild cognitive impairment. Over sixteen weeks, the lion's mane group improved on a cognitive test (Mori et al., 2009). In the wider literature, improvements of this kind have tended to fade once supplementation stopped, which points to a possible effect that depends on continued use rather than a lasting change. Research has also been done in clinical populations, which is a separate context from everyday use and not a basis for any consumer claim.
Cognition and mood in healthy adults
Interest has grown in whether lion's mane does anything for people who are not cognitively impaired. Small studies have looked at mood, focus and subjective cognitive performance in healthy adults (Nagano et al., 2010), and more recent placebo-controlled work has examined acute effects on cognition and mood. These studies are small, often rely on self-reported outcomes, and results across the wider literature are inconsistent.
The gut-brain axis
One of the more interesting recent directions is that lion's mane may act partly through the gut-brain axis. Its polysaccharides appear to influence gut microbiota, which in turn relate to inflammation, mood and cognition. This is an early-stage but promising area, and it is a more distinctive angle than the crowded "focus" claims.
What the science does not show
This section is deliberate, because it is where most supplement marketing goes wrong, and where regulators pay attention.
Lion's mane is not a proven treatment for anxiety or depression. Small studies have shown mood changes, but it has not been tested as a treatment for clinical mood disorders.
Improving a test score in mild cognitive impairment is not the same as reversing neurodegeneration. These are different things, and the evidence does not support the leap.
Lion's mane is not a stimulant and not an alternative to prescription medication. It works through entirely different mechanisms.
Animal studies showing nerve regeneration do not mean lion's mane "repairs" the human brain. Human evidence for that is absent.
Holding this line is not caution for its own sake. It is what the current evidence actually supports.
What science still needs to prove
Small samples. Most human studies involve fewer than fifty participants, which limits statistical power.
Short duration. Many trials run only four to sixteen weeks, too short to assess long-term effects.
Standardisation. Studies use whole fruiting body, mycelium, alcohol extracts, hot-water extracts and isolated compounds, which makes them hard to compare.
Clinical endpoints. Few studies use objective biomarkers or brain imaging.
Specific populations. More is needed on long-term use, use in pregnancy, and interactions with medications.
Is lion's mane safe?
Lion's mane has a long history of safe culinary use and is generally well tolerated as a supplement. In trials, the main issues were mild and digestive, such as abdominal discomfort or nausea, with occasional skin rash. Serious adverse events have not been reported in human studies to date, though rare individual case reports exist.
